You cannot do ISOs unless you have a DLNA server that will transcode them. The Hopper appears to play .mkv files just fine so you can convert them and they will play. I ripped a few SD DVDs to mkv and they worked with 5.1 sound.
There are several good free DLNA servers out there and Windows 7 has DLNA built into Windows Media Player.
