Does the HR10-250 support folders?

No, and I don't think there are any plans for it either.

The HR10-250 is running 3.1.x software, and thus, doesn't have grouping and folders built in.

DirecTV claims to be planning to upgrade them at some point -- however, there's little value in doing so, as far as they're concerned.

No, it doesn't. The Hughes box is sort of a stepchild, because TiVo does not use its Series 2 TiVo software in it, and most of the TiVo updates do not apply to DirecTV boxes at all. Since DirecTV appears to be abandoning TiVo, I think you are going to have be a regular TiVo subscriber to get all you want, because D* boxes will never have the latest cool TiVo stuff.
